Cet ouvrage rédigé entre 1856 et 1894 est un des plus importants en physiologie qui ait paru à cette époque. Cette étude se divise en trois parties bien distinctes : la dioptrique de l'oeil, les sensations visuelles, les perceptions correspondantes. Cette première partie donne les moyens par lesquels le mouvement vibratoire extérieur est conduit et transmis jusqu'à la surface nerveuse pour y produire une impression. Helmholtz y présente sa théorie de l'accommodation et une description de l'ophtalmoscope.
