About the Book

The breast cancer resistance protein (BCRP/ABCG2) transports drugs and carcinogens, some of which are substrates of cytochrome P450 1A1 (CYP1A1). Since transporters and enzymes are known to share similar substrates and regulatory pathways and evidence suggests that aryl hydrocarbon receptor (AHR) may also regulate transporters, it is hypothesized that various AHR agonists would induce BCRP levels in an AHR-dependent manner. In silico analysis of the 5'-flanking region of the human and mouse BCRP gene revealed 21 and 5 putative AHR response elements (AHREs), respectively. In vitro analysis in various human cell lines (colon, hepatocellular and mammary carcinoma and non-carcinoma) demonstrated an AHR-dependent induction of BCRP by AHR agonists in some cell lines. In vivo analysis revealed that 2,7,3,8-tetrachlorodibenzo-p-dioxin (a potent AHR agonist) does not induce mouse Bcrp1 levels in the brain, liver and intestine. Taken together, this suggests that the induction of BCRP levels by AHR agonists is cell-line and species-specific.
