We tested whether Pain1, a mouse quantitative trait locus (QTL) for neuropathic pain behaviour (autotomy), also underlies this behaviour in rats. Genotyping chromosome 7q34, the syntenic genomic region of Pain1, in rat lines expressing contrasting levels of autotomy, revealed linkage between markers in 7q34 and autotomy in some, but not all rat lines. Thus, Pain1 is relevant for autotomy in the rat but is not the only locus controlling this trait. Our second aim was testing whether autotomy in the mouse is controlled by one or several QTLs. Autotomy levels were determined in male and female offspring of reciprocal crosses between A/J, a high-expressing inbred mouse strain, and CBA/J, DBA/2J and C3H/HeJ, low-expressing inbred strains. We show that DBA and CBA contrast with A at one QTL for autotomy, whereas C3H contrasts at a second QTL. We conclude that autotomy levels in rodents is controlled by a number of genetic loci.
